Study of Actions of Prostaglandin E©ûon the Vas Deferens and Seminal Vesicle of Guinea-pig

Abstract
The author investigated the potentiating effects of PGE, on the actions of sympathomimetics, acetyl¡©choline and histamine in the isolated vas deferens, and seminal vesicle of the guinea-pig.
The results are as follows:
1. In the isolated guinea-pig vas deferens and seminal vesicle, the actions of epinephrine, phenyleph¡©rine and norepiinephrine were much potentiate3 by pretreatment with PGE,. Isoproterenol, which has no. action alone on the vas deferens and seminal vesicle, produced strong contraction after PGE, pretreatment and the contraction could be blocked by phenoxybenzamine.
2. The action of acetylcholine on the vas deferens and seminal vesicle was augmented by PGE, pre¡©treatment When the action of acetylcholine was blocked by its specific antagonist, atropine, PGE, could not reverse the block; however, when blocked by phenoxybenzamine, PGE, did eliminate the blockade.
3. The action of histamine on the vas deferens and seminal vesicle was also enhanced by PGE, pretreat¡©ment The action of histamine did not appear following PGE, when the preparation was treated with¡©pyribenzamine but did reappear with PGE, when the preparation was blocked by phenoxybenza¡©mine. Phentolamine did not affect the actions of acetylcholine or histamine, but completely blocked the action of epinephrine.
4. In the vas deferens of reserpinized guinea-pig, the actions of epinephrine, acetylcholine and hista¡©mine were potentiated; but in the seminal vesicle of this animal, the actions of acetylcholine and his¡©tamine were lessened by PGE, pretreatment
5. In the isolated hypogastric nerve-vasa preparation of guinea-pig, the contractile response to hypogas¡©tric nerve stimulation was also augmented by the administration of PGE, alone in the reserpinized or guanethidine-treated guinea-pig. This action was not completely blocked by pretreatment with phentolamine or atropine.
6. It might be assumed that PGE, induced a supersensitizstion by increasing excitability of several ceptors to agonist in the pastsynaptic membrane of the vas deferens and seminal vesicle of the guinea-pit
